Plasticity of non-shivering thermogenesis and brown adipose tissue in high-altitude deer mice

چکیده

ABSTRACT High altitude environments challenge small mammals with persistent low ambient temperatures that require high rates of aerobic heat production in face O2 availability. An important component thermogenic capacity rodents is non-shivering thermogenesis (NST) mediated by uncoupled mitochondrial respiration brown adipose tissue (BAT). NST plastic, and for increases cold acclimation. However, lowland native rodents, hypoxia inhibits BAT. We hypothesize highland deer mice (Peromyscus maniculatus) overcome the hypoxic inhibition through changes BAT function. tested this hypothesis using lab born raised mice, a congeneric leucopus), acclimated to either warm normoxia (25°C, 760 mmHg) or (5°C, 430 mmHg). determined effects acclimation ancestry on whole-animal NST, mass interscapular (iBAT), uncoupling protein (UCP)-1 expression. To identify function, we conducted high-resolution respirometry isolated iBAT mitochondria substrates inhibitors targeted UCP-1. found increased but only mice. There was no effect any group, showed UCP-1 expression UCP-1-stimulated response these stressors. Our results suggest have evolved increase chronic hypoxia, driven part

Brown adipose tissue and thermogenesis.

The growing understanding of adipose tissue as an important endocrine organ with multiple metabolic functions has directed the attention to the (patho)physiology of distinct fat depots. Brown adipose tissue (BAT), in contrast to bona fide white fat, can dissipate significant amounts of chemical energy through uncoupled respiration and heat production (thermogenesis). This process is mediated by...

Mechanisms of thermogenesis in brown adipose tissue.

In 1969, Rafael and colleagues observed that the uncoupled state of mitochondria prepared from the brown fat of newborn guinea pigs could be reversed by incubation in the presence of albumin and certain purine nucleotides (Rafael et al., 1969). In 1974, we showed that this uncoupled state was due to the presence in the inner membrane of the mitochondria of a regulatable ion conductance pathway,...

Central Control of Brown Adipose Tissue Thermogenesis

Thermogenesis, the production of heat energy, is an essential component of the homeostatic repertoire to maintain body temperature during the challenge of low environmental temperature and plays a key role in elevating body temperature during the febrile response to infection. Mitochondrial oxidation in brown adipose tissue (BAT) is a significant source of neurally regulated metabolic heat prod...

Hypothalamic control of brown adipose tissue thermogenesis

It has long been known, in large part from animal studies, that the control of brown adipose tissue (BAT) thermogenesis is insured by the central nervous system (CNS), which integrates several stimuli in order to control BAT activation through the sympathetic nervous system (SNS). SNS-mediated BAT activity is governed by diverse neurons found in brain structures involved in homeostatic regulati...
